Nature of human cerebral vascular pigment and its association with age,sex,and brain regions
Objective To investigate the nature of cerebral vascular pigment(CVP) in different individuals and its association with age,sex,and brain regions. Methods Brain tissue samples were collected from 146 forensic autopsy cases,and the nature of CVP was analyzed by HE staining,Prussian blue staining,modified Fouchet bile pigment staining,the alkylphenol ethoxylate(APE) method for removing formalin pigment,the potassium permanganate/oxalic acid(PPO) method for removing melanin,ferrous sulfate staining,Schmorl staining,and immunohistochemical staining of tyrosinase-related protein 1(TRP-1). SPSS 26.0 software was used for statistical analysis,and the chi-square test was used to analyze the detection of CVP across different sexes,ages,and brain regions. Results CVP appeared to be brown-yellow in HE staining,Prussian blue staining,modified Fouchet bile pigment staining,and APE staining,and it was successfully removed by PPO staining for removing melanin. CVP exhibited greenish-black changes in ferrous sulfate staining and Schmorl staining and showed red-positive results in TRP-1 immunohistochemical staining. There was a significant difference in the detection rate of CVP across different age groups(P<0.001),which aggravated with the increase in age;there was a significant difference in the detection rate of CVP between different brain regions(P<0.001),and the detection rate of CVP in the parietal lobe was significantly higher than that in the frontal lobe,the temporal lobe,and the occipital lobe; however,there was no significant difference in the detection rate of CVP between different sexes(P>0.050). Conclusion CVP is melanin pigment,and exploration of its potential sources and functions can provide a basis for nervous system-related disciplines and promote the development of neuroscience,especially neuropathology.
